Abstract:

Introduction: Ischemic Preconditioning (IPC) is the phenomen that happens on the heart by one or several short periods of ischemia followed by reperfusion that improve the postischemic recovery of mechanical function. Ischemic preconditioning (IPC) may protect the heart from ischemia reperfusion injury by nitric oxide formation. This study investigated the effect of ischemic preconditioning on heart and the relationship between nitric oxide. Methods: 28 male Sprague dawley rats (200-250 g) in Tehran University of Medical Sciences were used. Rats were anesthetized and hearts were rapidly isolated and perfused in the Langendorff mode at a constant perfusion pressure and temperature of 37 ̊C. hearts were divided to 4 groups. Control group was perfused 170 minutes with buffer solution. ischemic reperfusion (IR) group was subjected to 30 minutes ischemia. (Ischemic preconditioning) IPC group was elicited by 5 min ischemia followed 5 min reperfusion before IR and L-NAME + IPC group, L-NAME (0.1mM) was added into the perfusion solution. Heart rate (HR), left ventricular development (LVDP), RPP (LVDP × HR), infarct size and coronary flow (CF) were measured. ANOVA tests( with TUKEY post test if p<0.05) were used for statistical analyses. Results: IPC improve the postischemic recovery and reduced postischemic ventricular dysfunction in heart and reduction of infarction size. No significant differences were observed between IPC and IPC + L-NAME groups. Conclusion: L-NAME did not affect postischemic recovery of IPC so in the isolated heart NO isn't involved in the cardioprotective effect of IPC.

Effect of crocin on nitric oxide synthase expression in post-ischemic isolated rat heart

Objective: Oxidative stress damages cells and brings about the pathogenesis of ischemia/reperfusion injury. This study was carried out to investigate the preconditioning and cardio protective potential effects of crocin and vitamin E by the eNOS and iNOS express gene in ischemia/reperfusion in rats. Material & Methods: Male rats were divided into seven groups, namely: sham, control group and ex...

Protective effect of nitric oxide induced by ischemic preconditioning on reperfusion injury of rat liver graft.

AIM Ischemic preconditioning (IP) is a brief ischemic episode, which confers a state of protection against the subsequent long-term ischemia-reperfusion injuries. However, little is known regarding the use of IP before the sustained cold storage and liver transplantation. The present study was designed to evaluate the protective effect of IP on the long-term preservation of liver graft and the ...

Roles of the nitric oxide signaling pathway in cardiac ischemic preconditioning against myocardial ischemia-reperfusion injury

Nitric oxide (NO), a vasoactive gas that can freely diffuse into the cell, has many physiological effects in various cell types. Since 1986, numerous studies of ischemic preconditioning against ischemia-reperfusion (I/R) injury have been undertaken and the roles of the NO signaling pathway in cardioprotection have been explored. Many studies have confirmed the effect of NO and that its relative...

Role of Endogenous Vitamin E in Renal Ischemic Preconditioning Process: Differences between Male and Female Rats

Background: Antioxidants such as α-tocopherol (vitamin E) and β-carotene (vitamin A) play an important role in protective effect of repeated brief periods of ischemia, namely ischemic preconditioning (IPC). Values of these antioxidants were investigated and compared after induction of ischemia reperfusion (IR) and kidney IPC in both male and female rats. Methods: Forty eight Wistar rats were di...

Role of matrix metalloproteinase II on analgesic effect of nitric oxide inhibition in rat

Abstract Introduction: Matrix metalloproteinase 2 is one of the inflammatory mediators that is involved in nociceptive processing and its production is regulated by many inflammatory factors such as nitric oxide. We studied the role of MMP-2 on the analgesic effects of nNOS inhibitor. Methods: Considering that nitric oxide has many roles in pain processing, we studied the CSF levels of MMP-2 ...
